Talent scouts begin their search for the Last Comic Standing. The search begins in New York City with talent scouts Richard Belzer and Steve Schirripa. The search then moves on to Tempe with talent scouts Fred Willard and Kathy Najimy.
Last Comic Standing travels to Los Angeles and Houston to select who will make it to the semifinals. Guest talent scouts for this episode are Angela Kinsey and Oscar Nunez from The Office for the Los Angeles auditions. Neil Flynn and Alfonso Ribeiro judge the contestants in Houston. The six best comedians are chosen to advance to the next round.
The hunt for the Last Comic Standing continues with auditions. French Stewart and Josh Gomez scout for talent in San Francisco, while Dave Foley and Richard Kind scout for talent just north of the border in Toronto.

At the end, only 12 comedy acts will move on to the next round and continue to compete to be the Last Comic Standing.
The comics head into the boxing ring, where they will have to be quick on the comebacks, disses, zingers and so forth. After three rounds, the winner will receive immunity.
The comics get to go on shopping spree. However, it is not a typical shopping spree. The comics go on the spree to look for props that they can use in the next contest, where the comics will perform for people visiting and working at the store. Carrot Top will help judge the performances of the comics, as two comics will be eliminated.
Eight comics remain as they head to the Playboy mansion. There, the comics will have to tell some bedtime stories to some of the Playboy models. The Playboy models that will be told the bedtime stories are Holly, Bridget, and Kendra; however, the visit would not be complete without a visit from Hugh Hefner himself.
The final five comics are revealed in this episode. The irreverent group the Dan Band perform.
The season six winner is revealed, along with a special performance by Jon Lovitz and Jon Reep (season 5 winner), plus an appearance by Triumph the Insult Comic, Richard Belzer, Steve Schirippa and Joel McHale. Plus, funny ads for Barack Obama and John McCain.
