The once great sketch comedy show 'Studio 60' is suddenly close to cancellation due not only to its less than stellar on-air product, but another, more unexpected type of action, its Executive Producer's on-air mental meltdown. So enter NBS Chairman of the board Jack Rudolph and a new network president, looking to put her mark on the struggling network, Jordan McDeere. This results in the re-hiring of Danny Tripp and Matt Albie to replace the former Executive Producer and hopefully save the show.
The Studio 60 cast and crew continue to deal with the fallout of Wes' on-air breakdown, as well as adjusting to Matt and Danny's management. Harriet finds out that Matt is sleeping with Jeannie, a cast member and writer. Meanwhile, Jeannie stresses when a sketch she pitched doesn't go over as well as she hoped; and the team scrambles to find a musical guest when the lead singer for The White Stripes catches tonsillitis.
A network focus group comes to the set, sending a scare through the cast and crew that the initial spike in the ratings after Matt and Danny took charge were just a fluke.
After 90 seconds of questionable material airs, the crew scrambles to recover. Meanwhile, Matt has to come up with a response to a gift from one of Harriet's fans and Jordan looks over an article written about the show.
As the cast rehearses the upcoming episode featuring Lauren Graham and Sting, a reporter tries to land a story about Matt and Harriet past history together. Meanwhile, Jordan deals the the backlash after she passes on a new reality series.
At an aftershow party, Jordan, tipsy after a few drinks, makes friends with the cast of the show. In order to get Matt's mind off of Harriet, Danny decides to hire three beautiful women. Meanwhile, Cal encounters a mysterious man backstage and Simon campaigns for more black writers.
Members of the Studio 60 staff travel to Nevada to get Tom out of jail and make it back before Friday's taping. There they encounter a judge who isn't a fan of the show and his disdain for the show interferes with their plan to get Tom before taping starts.
Jack pleads with the local Nevada judge (John Goodman) to free Tom so he can make it back to the Friday night show. Meanwhile, tension heats up between Matt and Harriet on the set.
After the show's ratings begin to drop, Matt finds out that both Ron and Ricky are planning to leave Studio 60 - and bring the writing staff with them. Meanwhile, Harriet debates whether or not do do a lingerie spread in a popular magazine and Jordan and Danny stress over budget cuts.
A virus runs rampant though the cast and crew, but the show must press on despite the illnesses and a small writing staff. Meanwhile, Jordan delivers surprising news to Danny.
With Christmas nearing, as well as the annual holiday Studio 60 episode, Matt sets himself on a personal mission to spread holiday cheer at the studio. Meanwhile, Danny is forced to confront his true feelings for Jordan.
The cast returns to the studio after their Christmas break, ready to get the show back in swing for the new year. While the cast and crew prepare for their first show, Danny continues to pursue Jordan. Meanwhile, Matt tries to win a date with Harriet by bidding on her in an online auction. However, Matt doesn't realize that winning the date will be the beginning of the end for them. Elsewhere, Jordan meets Hallie, the new Vice President of Alternative programming, but their meeting is anything but perfect.

In the aftermath of Matt and Harriet's broken relationships, flashbacks tell the story of how Matt and Harriet met for the first time. Matt thinks back to his early days as a writer and how he and Luke competed over Harriet's affections. Meanwhile, Danny and Jordan's relationship continues to grow and Tom and Dylan stress over whether or not their sketch will survive the 'Friday night slaughter'.
Matt is stricken with a case of writer's block and stays up an entire Wednesday night trying to come up with new material. In addition to the stress of not being able to write, Matt must also contend with a new lawyer who is looking into a sexual harassment claim while Harriet's on the set of her new movie. Meanwhile, Jordan and Danny square off to see who'd make a better parent.

During a live taping of a show, Jordan experiences emergency pregnancy complications. While Jordan is taken to the hospital, Tom has to deal with an unfolding emergency of his own. Meanwhile, Matt's pill abuse is uncovered.

The long day's journey into night continues as we learn more, in flashback, about how Matt and Danny came to be fired. Jack tries to convince Simon to apologize for his statement to the press. Matt continues to explore paying a ransom to free Tom's brother over the objections of Captain Boyle. Harriet sits with and act as a distraction for Danny who is keeping vigil for Jordan at the hospital.
Danny, Jordan, Tom, and Simon's long day finally comes to an end. Meanwhile, Matt and Harriet have a new start in their relationship.
